Introduction: Bats, along with their ectoparasites, harbor a wide diversity of symbiotic and potential pathogenic bacteria. Despite the enormous diversity of bats (181 species), few studies aimed to investigate the bacterial microbiome of Brazilian chiropterans and associated ectoparasites. This study aimed to characterize the bacterial microbiome of non-hematophagous bats and associated Streblidae flies and Macronyssidae and Spinturnicidae mites in the state of Mato Grosso do Sul, midwestern Brazil. Methods: Oral and rectal swabs were collected from 30 bats (Artibeus lituratus [n = 13], Artibeus planirostris [n = 9], Eptesicus furinalis [n = 5], Carollia perspicillata [n = 2], and Platyrrhinus lineatus [n = 1]). In addition, a total of 58 mites (15 Macronyssidae and 43 Spinturnicidae) and 48 Streblidae bat flies were collected from the captured bats. After DNA extraction and purification, each sample's bacterial composition was analyzed with metagenomic sequencing. Results: The microbiome composition of both oral and rectal bat swab samples showed that Gammaproteobacteria was the most abundant bacterial class. Spiroplasma, Wolbachia and Bartonella represented the most abundant genera in Streblidae flies. While Wolbachia (Alphaproteobacteria) was the most abundant genus found in Spinturnicidae, Arsenophonus (Gammaproteobacteria) was found in high abundance in Macronyssidae mites. In addition to characterizing the microbiome of each sample at the class and genus taxonomic levels, we identified medically significant bacteria able to infect both animals and humans in oral (Streptococcus and Anaplasma) and rectal swabs (Enterobacter, Klebsiella, Escherichia, Enterococcus, Streptococcus), Macronyssidae (Anaplasma, Bartonella, Ehrlichia) and Spinturnicidae (Anaplasma, Bartonella) mites as well as Streblidae flies (Spiroplasma, Bartonella). Discussion and conclusion: Besides expanding the knowledge on the bacterial microbiome of non-hematophagous bats and Streblidae flies from Brazil, the present work showed, for the first time, the bacterial community of bat-associated Macronyssidae and Spinturnicidae mites.

Ectoparasites found on bats are known to contain important microbes. However, the viruses hosted by these obligate parasites are understudied. This has led to the near oversight of the potential role of these ectoparasites in virus maintenance and transmission from bats to other interacting species and the environment. Here, we sampled bat ectoparasites parasitizing a diverse selection of bat species in the families Rhinolophidae, Vespertilionidae, Megadermatidae, Hipposideridae and Pteropodidae in Yunnan Province, China. We show that the ectoparasite prevalence was generally higher in male compared to female bats. Most ectoparasites were found to fall within the Nycteribiidae, Spinturnicidae and Streblidae bat ectoparasite families. We subsequently applied a non-biased sequencing of libraries prepared from the pooled ectoparasites, followed by an in-silico virus-centric analysis of the resultant reads. We show that ectoparasites hosted by the sampled families of bats are found to carry, in addition to a diverse set of phages, vertebrate and insect viruses in the families Aliusviridae, Ascoviridae, Chuviridae, Circoviridae, Flaviviridae, Hepadnaviridae, Hepeviridae, Herpesviridae, Iridoviridae, Marseilleviridae, Nairoviridae, Orthomyxoviridae, Parvoviridae, Poxviridae, Reoviridae, Retroviridae, and Rhabdoviridae. We further report a partial Parvovirus VP1/VP2 gene and partial Poxvirus ubiquitin-like gene predicted by two independent next generation sequencing data analysis pipelines. This study describes the natural virome of bat ectoparasites, providing a platform for understanding the role these ectoparasites play in the maintenance and spread of viruses to other animals.

RESUMO

Periglischrus calcariflexus new species, was found on the Greater Long-tongued bat Leptonycteris nivalis (Saussure) in Mexico. The female, male, deutonymphs, and protonymph are described and illustrated. Additionally, we evaluate the morphological variability of this species based on its geographical distribution. Considering this new species, the number of species in the vargasi species group within the genus Periglischrus increases to five; therefore, herein we include a key for the known species in the vargasi species group for the Neotropics.

Schreiber's bent-winged bat Miniopterus schreibersii and the greater horseshoe bat Rhinolophus ferrumequinum are widespread and common cavernicolous species across southern Europe that host numerous specialized ectoparasite species. The objective of this study was to characterize the species assemblage, genetic diversity, and host specificity of bat flies (Nycteribiidae, Diptera) and wing mites (Spinturnicidae, Acari) found on these bat hosts in Serbia and Bosnia and Herzegovina. Notably, while bat flies lay puparia on the cave walls and can thus be transmitted indirectly, wing mites require direct body contact for transmission. Morphological identification and sequencing of a 710-bp fragment of cytochrome oxidase I gene of 207 bat flies yielded 4 species, 3 on M. schreibersii and 1 on R. ferrumequinum. Sequencing of a 460-bp small subunit ribosomal RNA fragment, in all 190 collected wing mites revealed 2 species, 1 per host. In no case was a parasite associated with 1 host found on the other host. Species and genetic diversity of flies were higher in M. schreibersii, likely reflecting their host's larger colony sizes and migratory potential. Mite species of both hosts showed similarly low diversity, likely due to their faster life history and lower winter survival. Our findings highlight a remarkably high host-specificity and segregation of ectoparasite species despite direct contact among their hosts in the roost, suggesting a defined host preference in the investigated ectoparasite species. Furthermore, the differences in ectoparasite genetic diversity exemplify the interplay between host and parasite life histories in shaping parasite population genetic structure.

We evaluated the morphometric variation of wing mite Periglischrus paracaligus Herrin and Tipton, along with the distribution of their host Leptonycteris yerbabuenae Martinez and Villa, in Mexico. A total of 115 female and 96 male specimens of P. paracaligus were used to conduct linear and geometric morphometric analyses. We assessed the influence of the geographic distribution of the migratory and nonmigratory populations of its bat host species on changes in size and shape on these parasites. Both analyses revealed high intraspecific variation in P. paracaligus, but subtle geographic differentiation. None of the approaches used identified a consistent pattern that separates unambiguously migratory from nonmigratory populations. Females presented more phenotypic variation than males and UPGMA analyses showed southern and northern colonies grouped in two distinct clades. Males on the other hand showed randomly grouped colonies with no geographic concordance. Interestingly, the most differentiated colony was the north Pacific colony of Jalisco. For both, males and females, isolation by distance (IBD) was not observed. We discuss these results as a possible scenario of contact between migratory populations located in northern Mexico with nonmigratory populations in other localities in central and southern Mexico conforming to a panmictic population along with their distribution range.

Bartonella species are facultative intracellular bacteria and recognized worldwide as emerging zoonotic pathogens. Bartonella were isolated or identified by polymerase chain reaction (PCR) in bats and their ectoparasites worldwide, whereas the association between them was scarce, especially in Asia. In this study, a retrospective analysis with frozen samples was carried out to identify the genetic diversity of Bartonella in bats and their ectoparasites and to investigate the relationships of Bartonella carried by bats and their ectoparasites. Bats and their ectoparasites (bat flies and bat mites) were collected from caves in Hubei Province, Central China, from May 2018 to July 2020. Bartonella were screened by PCR amplification and sequencing of three genes (gltA, rpoB, and ftsZ). Bats, bat flies, and bat mites carried diverse novel Bartonella genotypes with a high prevalence. The sharing of some Bartonella genotypes between bats and bat flies or bat mites indicated a potential role of bat flies and bat mites as vectors of bartonellae, while the higher genetic diversity of Bartonella in bat flies than that in bats might be due to the vertical transmission of this bacterium in bat flies. Therefore, bat flies might also act as reservoirs of Bartonella. In addition, human-pathogenic B. mayotimonesis was identified in both bats and their ectoparasites, which expanded our knowledge on the geographic distribution of this bacterium and suggested a potential bat origin with bat flies and bat mites playing important roles in the maintenance and transmission of Bartonella.

Abstract Present study was conducted to record ecotoparasitic prevalence in bat fauna of the northwestern parts of Pakistan. A total of 204 bat specimens representing 14 species were captured during a two year survey, extending from June 2015 through May 2016. A species of soft ticks Argas vespertilionis was identified from 23 bat specimens. Similarly, members of the family Dermanyssoidae (dermanyssoid mites) were isolated from 10 bat specimens, that of Spinturnicidae (spinturnicid mites) from 3 and Streblidae (bat flies) from 2 bat specimens. These parasites were collected using entomological tweezers and were identified on morphological basis. Further studies on parasitic prevalence, molecular characterization of bat parasites and their control measures are recommended.


Resumo O presente estudo foi realizado para registrar a prevalência de ectoparasitas na fauna de morcegos em partes do noroeste do Paquistão. Um total de 204 espécimes de morcegos, representando 14 espécies, foi capturado durante uma pesquisa de dois anos, de junho de 2015 a maio de 2016. A espécie de carrapato Argas vespertilionis foi identificada em 23 espécimes de morcegos. Da mesma forma, os membros da família Dermanyssidae (ácaros dermanyssoid) foram isolados de 10 espécimes de morcego, os da Spinturnicidae (ácaros spinturnicid), de 3, e os da Streblidae (mosca de morcego), de 2 espécimes de morcego. Esses parasitas foram coletados com pinça entomológica e identificados com base morfológica. Estudos adicionais sobre prevalência parasitária, caracterização molecular de parasitas de morcego e suas medidas de controle devem ser realizados.

The discovery of a new species, Periglischrus empheresotrichus, was determined through a review of museum collections, as well as a field survey of ectoparasites of island bats. This new species parasitizes on two bat species of the genus Monophyllus Leach, the Greater Antillean Long-tongued bat Monophyllus redmani Leach and the Lesser Antillean Long-tongued bat Monophyllus plethodon Miller. The female, male, deuthonymphs, and protonymph are described and illustrated. P. empheresotrichus n. sp. has an insular distribution, we evaluated the morphological variation of the adult populations, and concluded that intra-specific variation is correlated both with host species and locality (island) in the West Indies.

Hematophagous Spinturnix myoti mites and their host, the greater mouse-eared bat (Myotis myotis), were tested for the presence of Bartonella spp., Rickettsia spp., and Anaplasma phagocytophilum. In total, Bartonella spp. DNA was amplified in 28% of 134 mite pools and in 25% of 59 bats tested by PCR targeting a fragment of citrate synthase gltA gen. Adult mites were at least threefold more frequently infected compared to immature stages. The overall infection prevalence among mite pools from cave-dwelling bats was higher than for those collected from attic shelters. Three distinct genotypes were detected. The most prevalent genotype in mites and bats matched closely with Candidatus Bartonella hemsundetiensis identified in bats from Finland and was relatively distant from bat-borne Bartonella strains described in the UK and France. Importantly, most sequences were close to those reported in forest workers from Poland. The presence of identical genotype among S. myoti samples and M. myotis bats suggests that bartonellae can be shared between mites and their bat hosts. In this case, wing mites could serve as vectors, whereas their hosts as reservoirs. One blood sample was positive by PCR for the msp2 gene of A. phagocytophilum. Two mite pools yielded Rickettsia spp. DNA. Both sequences were distinct from any known species but can be classified as spotted fever group Rickettsia spp. Our findings expanded our knowledge on the role of spinturnicid mites in the ecology and epidemiology of bacterial infections associated with vespertilionid bats, especially regarding the genus Bartonella.

In Brazil, 124 species of ectoparasites of bats are known from the families Streblidae, Nycteribiidae, and Spinturnicidae. This study aimed to characterize the ectoparasites associated with bats in a restinga habitat in Sergipe, northeastern Brazil. Sampling occurred between October 2016 and September 2017 in the Reserva Particular do Patrimônio Natural do Caju through the capture of bats, using 10 mist nets. The ectoparasites collected were stored in 70% alcohol and subsequently identified. For the hosts, the parasitological indexes were calculated and, for the most abundant ones, the influences of the sex of the host and of seasonality on these indexes were evaluated. We collected 430 parasites, of which 77.90% belonged to the Streblidae family. The influence of host sex was observed only in terms of the prevalence between Periglischrus iheringi and Artibeus lituratus, with the highest values observed for females. This may be associated with the highest susceptibility of females to parasitism by the greater permanency in the shelter. In terms of the influence of seasonality, only the interaction between Carollia perspicillata and Trichobius joblingi was affected by temperature, with an increase in prevalence rates and mean intensity during the season with lower temperatures. This relationship may be associated with biological differences among parasites species. The present study identified six new species of ectoparasites and the occurrence of ticks on bats for Sergipe, in addition to registering 11 new bat-ectoparasite associations for the state and two for Brazil, contributing to an increase in our knowledge about bat-ectoparasite interactions.

Ectoparasites of bats in the Neotropics are diverse and play numerous ecological roles as vectors of microbial pathogens and endoparasites and as food sources for other cave fauna living both on their hosts and in bat roosts. The ectoparasites of bats in Jalisco State of western Mexico have not been as well described as those of other states with recent checklists that have focused primarily on the Yucatan Peninsula. We captured bats from 2011-2015 on the south coast and Sierra de Amula, Jalisco using mist nets, and we removed ectoparasites by hand. We identified 24 species of streblid bat flies and six ectoparasitic mites from bats caught in mist nets. There were an additional eight possibly undescribed species of Streblidae. Our collections extend the known range of species into Jalisco.

Ectoparasites of bats and bat-associated pathogens are poorly studied in the Lesser Antilles Islands. We report on an 11-mo field study on Saint Kitts Island of bat populations, their associated ectoparasites, and pathogens. We report on five ectoparasite species, including four Streblidae (Diptera) and a Spinturnicidae (Acari). Several genotypes of unnamed Bartonella were isolated from bats and ectoparasites. Microfilaria of an undetermined Litomosoides spp. were detected in blood from Artibeus jamaicensis Leach (Chiroptera: Phyllostomidae) (and associated ectoparasites: Trichobius intermedius Peterson and Hurka (Diptera: Streblidae) and Periglischrus iheringi Oudemans (Acari: Spinturnicidae)). In addition, an Ehrlichia sp. and Rickettsia africae were detected in the blood of several bat species. Our study is one of the first surveys of ectoparasite-borne pathogens in wild mammals from St. Kitts.
